This study aimed to extract and purify the papain enzyme from papaya leaves using two extraction methods - grinding and ultrasonication. Papain concentration was determined from the extracts and its proteolytic activity was assessed. Phytochemical analysis of the leaves revealed proteins and alkaloids to be most abundant. The maximum papain concentration of 0.054 mg/ml was obtained from grinding extraction while ultrasonication yielded a minimum of 0.002 mg/ml.
