What is Polymorphisms in inflammatory cytokines and Fcγ receptors in childhood chronic immune thrombocytopenic purpura: a pilot study about?

Inflammatory cytokines and low‐affinity Fcγ receptor (FcγR) polymorphisms were investigated in 37 children with chronic immune thrombocytopenic purpura (cITP) and 218 controls. Genotype analysis included common variants in the regulatory regions of cytokines, __TNF__, __LTA__, __IL1RN__, __IL1A__, __IL1B__, __IL4, IL6__ and __IL10__, and structural variants of the low affinity FcγRs, __FCGR2A__, __FCGR3A__ and __FCGR3B__. Associations were observed for __TNF__ (__P__ = 0·0032), __LTA__ (__P__ = 0·019), __FCGR3A__ (__P__ = 0·038) and __FCGR3B__ (__P__ = 0·0034). Two combinations of genotypes (__TNF__ and __FCGR3A;____P__ = 0·0003, and __LTA__ and __FCGR3B;____P__ = 0·011) were significantly associated with cITP. These results provide preliminary evidence that variant genotypes of FcγRs and cytokines contribute to cITP pathogenesis.
