The term narrow therapeutic ratio is defined in the US Code of Federal Regulations at 21 CFR 320.33(c) as follows 1 : (1) there is less than a 2-fold difference in median lethal dose (LD 50 ) and median effective dose (ED 50 ) values, or (2) there is less than a 2-fold differ-ence in the minimum toxic concentrations and minimum effective concentrations in the blood, and (3) safe and effective use of the drug products requires careful titration and patient monitoring.
