RATIONALE: Apoptotic cells are removed by phagocytosis, namely, efferocytosis. Efferocytosis is physiological removal of apoptotic cells, preventing the release of proinflammatory intracellular contents. Impairment of efferocytosis was reported in alveolar macrophages (AMs) of patients with COPD. In addition, histone deacetylase (HDAC) activity is reduced in AMs of COPD. Other studies reported that a HDAC inhibitor, trichostatin A (TSA), decreased CD9 expression on macrophages. CD9, a member of the tetraspan transmembrane proteins, facilitates uptake of virus-infected cells, and monocyte fusion. The regulation of efferocytosis is controlled by the Rho family GTPases, including Rac. These studies imply that status of HDAC activity and CD9 expression might link to efferocytosis. We aimed to investigate possible roles of HDAC and CD9 in efferocytosis. METHODS: Murine AMs were collected by bronchoalveolar lavage. Apoptotic cells were prepared by ultraviolet irradiation to the human polymorphonuclear leucocytes (PMNs).
