What is Antagonistic effect of CRP and KdgR in the transcription control of the Erwinia chrysanthemi pectinolysis genes about?
The main virulence factors of the phytopathogenic bacteria __Erwinia chrysanthemi__ are pectinases that cleave pectin, a major constituent of the plant cell wall. The cyclic AMP receptor protein (CRP) was identified as the main activator of the pectinolysis genes. Gel shift and DNase I footprinting experiments showed that the purified __E. chrysanthemi__ CRP protein binds specifically to the promoter regions of seven pectinolysis genes (__pelB__, __pelC__, __pelD__, __pelE__, __ogl__, __kduI__ and __kdgT__ ) whose expression is positively regulated __in vivo__ by CRP. In contrast, no interaction was observed between CRP and the promoter–operator region of __pelA__, whose expression is negatively regulated __in vivo__ by CRP. Primer extension experiments demonstrated that each of the __pelB__, __pelC__, __pelE__ and __kduI__ genes is expressed from a unique σ^70^ promoter, whereas __ogl__ and __kdgT__ possess three and two functional promoters respectively. The position of the CRP binding site relative to the transcription start site suggests that CRP acts as a primary activator at the __pelB__ (via the CRP binding site 1), __pelC__, __pelE__, __pelD__, __kdgT__ P~1~ and __ogl__P~2~
