Basal cell carcinoma (BCC) is the most common cutaneous malignancy and is characterized by sustained Hedgehog (Hh) signaling. Despite major insights into the molecular basis of BCC, relatively little is known about how BCC tumor cells affect other cell types in the tumor microenvironment. It has been frequently observed that a subset of BCC tumors are clinically pigmented. Using immunostaining for melanocyte-specific antigens, we found that most human and mouse BCCs contain tumor nodules with abundant proliferating melanocytes, even in tumors that are not grossly pigmented. We performed single-cell RNA sequencing of mouse BCCs to search for potential tumor-cell derived factors capable of driving melanocyte proliferation. We found that expression of the potent melanocyte mitogens Endothelins 1 and 2 (Edn1, Edn2) was markedly upregulated in BCC tumor cells, whereas the corresponding Endothelin receptor type B (Ednrb) was largely restricted to melanocytes.
