Phosphorylation is a central mechanism of cell regulation, and the dynamic interplay between kinases and phosphatases determines the phosphorylation state of a protein (2 or 3) within living systems. The caged phosphoprotein or peptide 1 gives researchers spatial and temporal control over the release of the phosphorylated species 2, yielding a new tool to elucidate its real time functional role. Two synthetic methods for caged phosphopeptides have been developed [1], and nonsense codon suppression has been adapted to incorporate caged phospho-amino acids into full length protein [2]. A validation study demonstrates the utility of these tools where classic genetic approaches cannot provide information [3].
