What is DNA Extraction and Isolation Techniques about?
DNA extraction involves four basic steps: 1) breaking open cells to expose the DNA, 2) removing membrane lipids with detergent, 3) optionally removing proteins with protease, and 4) precipitating the DNA using alcohol which causes it to aggregate and form a pellet. Proteins bound to the DNA can be removed by protease or by precipitating or extracting them prior to the DNA precipitation step. The purified DNA can then be dissolved in a slightly alkaline buffer or water.
