What is Gut Microbiome and Autism Spectrum Disorder about?

Maralixibat relieves chronic cholestasis in children Maralixibat is an apical sodium-dependent bile acid transport (ASBT) inhibitor that blocks enterohepatic recirculation of bile acids and could prolong transplant-free survival in children with chronic cholestasis due to Alagille syndrome. In ICONIC, a phase IIb placebo-controlled randomizedwithdrawal study, 31 children with Alagille syndrome received 380 μg/kg maralixibat once daily for 18 weeks. Of these, 29 children were randomly assigned to placebo or maralixibat for a further 4 weeks; the placebo-treated children experienced rising serum bile acid levels and worsening pruritus, which contrasted with sustained benefits in those who continued maralixibat. The 15 children who completed a 26-week open-label extension showed clinically meaningful improvements from baseline in serum bile acid levels, pruritus, xanthomas, height and quality of life. Maralixibat doses of up to 380 μg/kg twice daily were generally well tolerated. ASBT inhibitors could be an important treatment for cholestasis in Alagille syndrome.
