This study investigated the impact of two different methods for preparing agar growth media - autoclaving phosphate and agar together (PT method) versus separately (PS method) - on cultivating microorganisms. Using a pure culture of Gemmatimonas aurantiaca and environmental samples, significantly higher colony counts were observed on PS medium compared to PT medium.
