What is Understanding DNA Mismatch Repair: the Work of Paul L. Modrich about?
Modrich is probably best known for his work on DNA mismatch repair, a process that rectifies base-pairing errors that arise during replication and recombination. Using the methyl-directed pathway for processing DNA biosynthetic errors in Escherichia coli, Modrich made many fundamental contributions to the understanding of the repair process. Mismatch repair relies on signals within the helix to direct repair to the newly synthesized strand of DNA. In the methyl-directed pathway, newly synthesized DNA is transiently unmethylated at GATC sites, and it is the absence of this modification that directs repair to the new strand. By the early 1990s, Modrich had reconstituted the E. coli methyl-directed reaction in a purified system consisting of MutH, MutL, and MutS proteins, DNA helicase II, single strand DNA-binding protein, DNA polymerase III holoenzyme, several single strand-specific exonucleases, DNA ligase, ATP, and the four deoxynucleoside triphosphates (1). He determined that MutH, MutL, and MutS were responsible for interactions with the two DNA sites involved in the methyl-directed repair event. MutS bound to mismatched base pairs in a reaction independent of the presence of a d
