What is Whole Genome Amplification Permits the Detection of Circulating Tumor-derived K-Ras Mutations in the Plasma of Patients Treated with Radiation Therapy about?
Purpose/Objective(s): The ability to identify sensitive and specific blood-based biomarkers could enable oncologists to monitor the extent of tumor eradication during therapy and to identify disease recurrence following treatment. Tumors release genomic DNA into the circulation during therapy through cellular necrosis and apoptosis. Tumor DNA may be distinguished from normal circulating DNA by the identification of tumor-specific mutations. We present preliminary data demonstrating the ability to detect circulating DNA from the tumors of patients receiving combined chemotherapy and radiation. Materials/Methods: Patients undergoing pre-operative or definitive treatment for pancreatic, rectal, esophageal or non-small lung cancer had blood drawn prior to initiating therapy, at weekly intervals during therapy and following the completion of therapy. DNA was extracted from tumor tissue obtained during surgery from the same patients. For identification of K-ras mutations in surgical samples or in plasma, a sensitive restriction enzyme mediated method (REMS-PCR) was employed, followed by direct sequencing for identification of mutations. As standard DNA extraction procedures provide only
