What is Molecular Aspects of Anti-Cancer Drug Action about?

For example, this chapter contains the first full review of the development of substituted anthraquinones modelled on Adriamycin, yet already two members of this group are undergoing clinical trial. Adriamycin and daunomycin have long been known to have an effect on nuclear nucleic acid, and this is regarded as one of their major cytotoxic effects, if not the major effect (for reviews see Brown, 1978;Arcamone, 1978Arcamone, , 1981)). Consequently, most in vitro studies of analogues have involved examination of their interaction with DNA or their inhibition of cell-free nucleic acid synthesis. Most cell culture studies have considered uptake of drug, and/or cellular localisation of drug and/or effect on cellular nucleic acid synthesis. Some in vivo studies have given evidence of the effect on the nucleus. From consideration of all these studies there is a composite picture emerging of the relationship between structural changes in the anthracyclines and differences in molecular action. However, the natural anthracyclines are such an extensive group that, before considering these molecular actions of natural and semisynthetic anthracyclines, their biogenetic origins will be briefly o
