This paper describes a new method called EXPLORs for loading soluble proteins into exosomes in a light-controlled and reversible manner. EXPLORs integrate a photoreversible protein-protein interaction module with the endogenous process of exosome biogenesis to actively load cargo proteins. Treatment with protein-loaded EXPLORs significantly increases intracellular levels and function of cargo proteins in recipient cells in vitro and in vivo, demonstrating their potential for efficient intracellular delivery
