It is likely that the aggregation is the result of a covalent modification of this protein (61, although no specific differences in the covalent structures of the proteins have yet been reported. In order to perform a comparative analysis of the amyloid and the pre-amyloid protein, we developed a new purification scheme which allows the isolation of both proteins by an almost identical procedure. Microsomal fractions were prepared from normal or scrapie hamster brain homogenates by differential centrifugatlon, subsequently extracted with Triton X-114 and separated into detergent-rich and aqueous phases.
