This document provides instructions for isolating plant DNA using the CTAB method. CTAB buffer is used to disrupt plant cell membranes and allow access to nuclear material without degrading the DNA. Tissue is first ground to break open cells, then resuspended in CTAB buffer and incubated to lyse the cells. Chloroform is added to separate contaminants from the nucleic acids, and isopropanol precipitates the DNA. The DNA is then washed, dried, and resuspended for storage.
