The document describes an investigation of using the HaloTag (HT) protein fusion system to study protein function in the malaria parasite Plasmodium berghei. Transgenic P. berghei lines were generated expressing GFP fused to either wild-type HT or a mutant HT. Weak GFP fluorescence was detected in the transgenic parasite lines by flow cytometry and microscopy, indicating HT can be expressed as a fusion protein in P. berghei, although it may interfere with fusion protein function and stability.
