What is Contrast-enhanced Destruction Replenishment Ultrasound for Monitoring Angiogenesis in a Rat Tumor Model about?

Objective: Changes in ultrasound backscatter characteristics following therapeutically induced tissue damage have been noted for many years. Recent work suggests that a component of tissue response to injury, apoptosis (programmed cell death), is related to specific changes in ultrasound scattering at high frequency. In this study, we examine changes in the high-frequency backscatter power spectral characteristics of an experimental human tumor xenograft model in response to tissue injury to evaluate the specificity of ultrasound spectral changes. Methods: A human uveal melanoma (C918) xenograft model in BALB/c nu/nu mice was used. Anesthetized mice were scanned in serial sections with a two-axis linear stepper motor scanning system with a broadband high-frequency transducer and appropriate digitization. Following the pretreatment scan, animals were either killed with euthanasia solution; injected with TRAIL to induce apoptosis; treated with short-term hyperthermia (ϳ45°C/5 minutes); or treated with sub-threshold HIFU. Animals were scanned every half-hour up to 8 hours post-treatment. Select tumors were processed for histology and immunohistology including modified TUNEL and Anexin
