The WSG-ADAPT HER2+/HR- phase II trial evaluated the efficacy and safety of 12 weeks of neoadjuvant dual blockade with trastuzumab and pertuzumab, with or without weekly paclitaxel, in patients with HER2-positive early breast cancer. Results showed that the addition of paclitaxel significantly increased the pathological complete response (pCR) rate from 36.3% to 90.5% in early responders. The study concluded that early non-response to dual blockade is a strong predictor of failure to achieve pCR.
