What is Centrifugation: Basic principle, types about?
Study of biological molecules, such as subcellular organelles, macromolecules (proteins and nucleic acids), viruses, ribonucleoproteins and lipoproteins, etc., is of prime importance for studying the molecular aspects of the cells. For studying the properties of these subcellular structures, it is important to preserve their functions and properties during the process of their separation. One of the key techniques used for this purpose is centrifugation. Centrifugal forces generated during the centrifugation process are used in separation of subcellular components present in a tissue homogenate. Generally, there are three main methods (differential, rate-zonal and isopycnic centrifugation) of separation. Separation in case of differential and rate-zonal methods is a function of their size; however, difference in density of separating particles is employed in isopycnic centrifugation. Nature of separating particles determines the method of choice for their separation. In this chapter, we have covered the detailed study of different centrifugation techniques, their principles, applications, and safety protocols associated with their use.
