What is Effect Of Comt Genetic Background And Stress On Pain Following Tibial Fracture about?
Our previous data has shown that pharmacologic or genetic inhibition of COMT (catechol-o-methyltransferase) in rodents produces multi-site body pain that is enhanced after stress. Here, we sought to examine the effects of low COMT activity genotype and stress on pain following tibial fracture. COMT+/- or WT mice were randomly assigned to 1 of 4 groups: 1) sham stress + sham fracture 2) swim stress + sham fracture 3) sham stress + tibial fracture, or 4) swim stress + tibial fracture (N=3-5M/F). Mechanical hypersensitivity at plantar and abdominal sites was measured using von Frey filaments and a spinal needle, respectively. Depressive-like behavior was measured using the tail suspension test. In wild type mice, compared to the sham-sham group, the stress-sham, sham-fracture, and stress-fracture groups exhibited mechanical hyperalgesia on the ipsilateral paw ∼1-3 months. Further, among wild type females but not males, both stress groups exhibited hyperalgesia at remote abdominal sites during this 1-3 month time period. The wildtype stress-fracture group also exhibited depressive-like behavior evidenced by increased immobility time during the tail suspension test. In COMT+/- mice, the
