What is Blood Transcriptomics Based Biomarker for Prediction of Clinical Outcome in Locally Advanced Pancreatic Cancer Patients Treated with Concurrent IMRT, Gemcitabine and Cetuximab about?
Purpose/Objective(s): The presence of lymph node (LN) metastases is an important prognostic factor in patients who undergo PD for pancreatic cancer (PC). Retrospective studies suggest that higher number of TNE is associated with improved survival after PD +/À CRT. Extended lymphadenectomy has been proposed in order to improve pathologic staging and survival, although there is no conclusive evidence that this improves survival. We examined the impact of TNE and NPN on survival for patients treated with PD and CRT on RTOG 97-04, a phase III intergroup study. Materials/Methods: The effects of TNE and NPN on disease-free and overall survival (DFS, OS) were evaluated using univariate and multivariate analyses. TNE and NPN were analyzed as continuous variables. Multivariate Cox proportional hazards models were performed and included these variables: NPN, TNE, age, pathological tumor stage, surgical margin status, and treatment arm. NPN was also analyzed separately as a discrete variable (0 vs. 1-3 vs. .3) in univariate analyses of DFS and OS. For each distinct NPN category, actuarial estimates of OS and DFS were calculated using Kaplan-Meier methods. Results: A total of 538 patients were
